ABSTRACT
Abstract The in vitro derivation of human germ cells has attracted interest the last years, but their direct conversion from somatic not yet been reported. Here we tested ability male to directly convert into a meiotic cell-like phenotype by inducing them with combination selected key cell developmental factors. We started pool 12 candidates that were reduced 6, demonstrating ectopic expression line-related genes PRDM1, PRDM14, LIN28A, DAZL, VASA and SYCP3 induced (hFSK (46, XY) hMSC XY)) . Induced showed marked switch transcriptomic profile expressed several post-meiotic line related markers, progression, evidence epigenetic reprogramming approximately 1% able complete meiosis as demonstrated haploid status markers. Furthermore, xenotransplantation assays subset properly colonize spermatogonial niche. Knowledge obtained this work can be used create models study gamete-related diseases humans.
