Abstract Recent studies have demonstrated microRNAs (miRNAs) and proteins are beneficial to axon regeneration, which may be involved in Electroacupuncture (EA) therapy against stroke. In this study, we aimed determine the pivotal role of PirB EA-produced rehabilitation ischemic stroke; screen investigate potential miRNAs directly regulating expression. The results showed EA treatment enhanced regeneration new projections from corticospinal tract at 28 d after cerebral reperfusion injury rats. Then, found decreased pirb mRNA protein expression penumbra within days reperfusion. reduction facilitated neurite outgrowth oxygen-glucose deprivation injury. miRNA microarray level twenty kinds changed administration. bioinformatics study luciferase assay verified miR-181b regulated increased levels penumbras, improved neurobehavioral function through direct targeting regulate PirB, RhoA GAP43. conclusion, provide first evidence that enhances stroke by epigenetic changes act on its targets, such as miR-181b/PirB/RhoA/GAP43 axis, is a novel mechanism therapy.
