ABSTRACT
Abstract A chemically-activatable alkynyl steroid analogue probe has been synthesized for visualizing the lipid raft membrane domains by Raman microscopy. The probe, in which ring of its backbone is replaced with an group, was designed to enable activation alkyne signal through Eschenmoser-Tanabe fragmentation reaction oxidized cholesterol precursor bilayer membranes. observed form liquid-ordered raft-like on a model giant-liposome system similar manner as cholesterol, and large accumulated at 2120 cm −1 mapped microdomains microscope. moiety confirmed be converted from α,β-epoxy ketone group p -toluensulfonyl hydrazine under mild condition. Through reaction, activated liposomes. Furthermore, also detected living cells stimulated scattering ring-A-opened analogue, thus, provides first promising tool potentially unravelling intracellular formation trafficking cholesterol-rich microdomains.
