ABSTRACT
It is important to quantify the distribution of behavior amongst a population individual cells reach more complete quantitative understanding cellular processes. Improved high-throughput analysis single cell requires uniform conditions for with controllable cell–cell interactions, including diffusible and contact elements. Uniform arrays static culture adherent have previously been constructed using protein micropatterning techniques but lack ability control secretions. Here we present microfluidic-based dynamic array that allows both arrayed fluid perfusion environments cells. In our device no surface modification required loading done in less than 30 seconds. The consists physical U-shaped hydrodynamic trapping structures geometries are biased trap only HeLa were shown adhere at similar rate as on glass substrate. Additionally, rates death division comparable experiment. Approximately 100 isolated observed growing adhering field view spanning ∼1 mm2 greater 85% maintained within primary site after 24 hours. Also, 90% 5% had undergone apoptosis hours array. We anticipate uses drug toxicity physiologically relevant perfused dosages well investigation signaling pathways systems biology.
