ABSTRACT
A novel integrated continuous-flow microfluidic system was designed and fabricated for solid phase peptide synthesis (SPPS) using conventional reactants. The composed of a glass-based radial reaction chip, diffluent amino acid feeding reservoirs reagent pathways. tri-row cofferdam-fence structure supports trapping. Highly cross-linked, porous high-loading 4-(hydroxymethyl)phenoxymethyl polystyrene (HMP) beads were prepared SPPS. transfer losses, hazardous handling time-consuming processes in traditional cleavage steps avoided by being replaced with the on-chip treatment. Six peptides from an antibody affinity library against β-endorphin different lengths sequences obtained simultaneously on constructed within short time. This is automatic, integrated, effective, low-cost, recyclable environment-friendly not only SPPS but also other chemical syntheses.
