ABSTRACT
Qualitative and quantitative studies of glycosylation patterns various biologically important proteins represent a key field for the understanding their complex structure-function relationships. However, analysis glycoprotein glycans is usually undermined by tedious sample processing steps prior to detection, including deproteination, desalting removal some other non-glycan impurities, which results in considerable loss increased difficulty analysis. Herein we report facile versatile method isolation reducing from samples using sulfonyl hydrazine-functionalized polystyrene (SHPS) beads, namely SHPS-based glycan capturing procedure. This allows chemoselective efficient condensation aldehyde group with active hydrazine SHPS beads under anhydrous conditions, resulting formation hydrazone conjugates. The components samples, such as proteins, salts can be completely removed washing Regeneration performed via mild hydrolysis washed procedure compatible almost all current techniques derivatization or detection glycans. We have obtained essential data this method, optimized reaction linearity reproducibility quantitation, well final recovery ratio. Moreover, mass spectrometric biological milk, human blood plasma fetal bovine serum, was achieved, demonstrating good applicability novel
