ABSTRACT
Glycosylation is involved in several biological processes, and its alterations can reflect the process of certain diseases. Type 2 diabetes mellitus (T2DM) has attained status a global pandemic; however, difference salivary protein glycosylation between healthy subjects patients with T2DM not been fully understood. In present study, specimens from (n = 72) volunteers (HVs, n 80) were enrolled divided into discovery validation cohorts. A method combining lectin microarray blotting was employed to investigate confirm altered glycopatterns glycoproteins. Then, lectin-mediated affinity capture glycoproteins MALDI-TOF/TOF-MS performed obtain precise structural information glycans. As result, recognized by 5 lectins (LEL, VVA, Jacalin, RCA120 DSA) showed significant alteration saliva patients. Notably, glycopattern Galβ-1,4GlcNAc (LacNAc) LEL exhibited increase compared HVs both The results indicated that there 10 7 LacNAc-containing N/O-glycans (e.g. m/z 1647.586, 11 688.613 1562.470) identified only Besides, relative abundance 3 N-glycans O-glycans an These LacNAc increased patients, may contribute this alteration. Our findings provide useful understand complex physiological changes
