ABSTRACT
S-nitrosothiols are products of nitric oxide (NO) metabolism that have been implicated in a plethora signalling processes. However, mechanisms S-nitrosothiol formation biological systems uncertain, and no efficient protein-mediated process has identified. Recently, we observed ferric cytochrome c can promote S-nitrosoglutathione from NO glutathione by acting as an electron acceptor under anaerobic conditions. In the present study, show this mechanism is also robust oxygenated conditions, protein S-nitrosation via transnitrosation reaction cell lysate depleted exhibits lower capacity to synthesize S-nitrosothiols. Importantly, demonstrate functional living cells. Lower synthesis activity, donor synthase-generated NO, was found c-deficient mouse embryonic cells compared with wild-type controls. Taken together, these data point mediator This most concerted reported so far.
