ABSTRACT
The dried stem bark of Berberis kansuensis is a commonly used Tibetan herbal medicine for the treatment diabetes. Its main chemical components are alkaloids, such as berberine, magnoflorine and jatrorrhizine. However, role gut microbiota in vivo metabolism these has not been fully elucidated. In this study, an ultra-high performance liquid chromatography method coupled with Orbitrap mass spectrometry (UHPLC-Orbitrap-MS) technology was applied to detect identify prototype metabolites rat intestinal contents serum samples after oral administration B. extract. A total 16 40 were identified. primary metabolic pathways from extract demethylation, desaturation, deglycosylation, reduction, hydroxylation, other conjugation reactions including sulfation, glucuronidation, glycosidation, methylation. By comparing differences between diabetic pseudo-germ-free rats, we found that transformation some bufotenin, ferulic acid 4-O-β-D-glucopyranoside, magnoflorine, 8-oxyberberine, affected by microbiota. results revealed can affect constituents These findings enhance our understanding active ingredients key on them.
