ABSTRACT
Gouty arthritis (GA) is an inflammatory triggered by the deposition of monosodium urate monohydrate (MSU) crystals, causing pain, inflammation, and joint damage. Several drugs are currently employed to manage acute flares GA, but they either have limited effectiveness or induce severe adverse reactions. Ouratea spectabilis traditionally used in Brazil treat gastric ulcers rheumatism. The ethanolic extract O. stems (OSpC) four biflavanones (ouratein A - D) isolated thereof were evaluated a murine model GA induced injection MSU crystals. underlying mechanism action ouratein D was investigated vitro cell cultures measurement IL-1β levels ELISA Western blot analysis. administration OSpC (10, 30 100 mg/Kg, p. o.) reduced migration total cells, monocytes, neutrophils diminished CXCL1 synovial tissue. Among tested compounds, only (1 mg/Kg) cells it shown be active up 0.01 mg/Kg (equivalent 0.34 nM/Kg, o.). Treatment pre-stimulated THP-1 (differentiated into macrophages) BMDMs with release both macrophage lines. This biflavanone activation caspase-1 (showed increase cleaved form) supernatants cultured BMDMs, evidencing its modulating inflammasome pathway. obtained results demonstrate anti-gout properties point out as bioactive component assayed extract.
