ABSTRACT
We have investigated whether GH treatment influences the expression of UCP1, 2 and 3 mRNA in a KK-Ay obese mouse model. mice (n = 10) C57Bl/6J control were injected subcutaneously with human (1.0 mg/kg/day 3.5 mg/kg/day) for 10 days, compared physical saline. The weighed significantly less (p < 0.01 : 1.0 mg/kg/day, p 0.05 had smaller inguinal subcutaneous perimetric white adipose tissue (WAT) pads mg/kg/day), but increased skeletal muscle weight 0.05). brown (BAT) did not change significantly. Not only plasma free fatty acid glucose levels also insulin decreased. reduced HOMA-IR (homeostasis model assessment-insulin resistance) values suggested that resistance was improved by treatment. UCP1 after mg 2.8-fold vs. saline controls) 2.0-fold 1 treatment) BAT, 6.0-fold WAT controls). UCP2 2.2-fold control) 2.1-fold muscle. One administration stimulated 2.5-fold UCP3 On other hand, lean showed no significant difference body composition or parameters. show any GH. conclude UCP These findings suggest GH-induced thermogenesis may contribute to reduction energy expenditure.
