ABSTRACT
Bone metastasis microenvironment-related growth factors such as insulin-like factor 1 (IGF-1), transforming beta (TGF-β1), basic fibroblast (bFGF) and interleukin 6 (IL-6) show survival activity, thereby inhibiting chemotherapy-induced apoptosis of PC-3 prostate cancer cells in vitro. Recently, zoledronic acid has been shown to induce while overexpression parathyroid hormone-related protein (PTHrP) inhibits serum deprivation-induced cells. Consequently, we have investigated whether IGF-1, TGF-β1, bFGF, IL-6, and/or dexamethasone affect the expression PTHrP type I PTH/PTHrP receptor (PTH.1R) using relative quantitative PCR real-time (expression at mRNA level) immunocytochemical immunofluorescence analysis level). Our data that bFGF IL-6 increase its perinuclear localization, (50 μM, 100 μM for 24 h 48 h) suppress We did not detect any appreciable change PTH.1R due TGF- β1, or Therefore, it is conceivable bone factor/anti-apoptotic activity anticancer action/pro-apoptotic on mediated, least part, by differential modulation expression.
