ABSTRACT
The objective of this study was to investigate the dose-dependent effect 1α,25-(OH)2VD3 (Vit D3) on invitro proliferation goat luteinised granulosa cells (LGCs) and determine underlying mechanisms its action by overexpressing silencing vitamin D receptor (VDR) in LGCs. Results showed that VDR prominently localised GCs theca (TCs) expression increased with follicle diameter, but lower atretic follicles than healthy follicles. rate LGCs significantly higher Vit D3-treated groups control group, highest observed 10nM group; accompanied changes cell cycle-related genes. These data indicate D3 affects LGC a manner. Contrary knockdown effects, overexpression upregulated downregulated cycle- apoptosis-related genes respectively; moreover, supplementation enhanced these effects. results suggest patterns may be associated follicular development regulation apoptosis. findings will enhance understanding roles ovarian development.
