ABSTRACT
Oocyte in vitro maturation (IVM) is crucial for subsequent embryo production. It involves acquisition of competence fertilization and development. Therefore, its optimization could have a direct impact on Dimethyl sulfoxide (DMSO) commonly used as solvent or vehicle, but also increases the membrane permeability behaves scavenger cytotoxic free radicals. The aim this study was to evaluate effect DMSO supplementation during bovine oocyte development determine optimal usage dose with no toxic effect. To aim, cumulus-oocyte complexes were collected from slaughterhouse ovaries IVM TCM 199 containing 10% fetal serum, 10 µg mL−1 FSH, 0.3 mM sodium pyruvate, 100 cysteamine, 2% antibiotic-antimycotic. oocytes incubated 24 h at 6.5% CO2 humidified air 38.5°C. For Experiment 1, medium supplemented concentrations 0, 0.1, 0.5, (vol/vol) (n = 241, 195, 42, 192, 172 oocytes) rate determined by presence first polar body. 2, 0.25, 0.75, 446, 322, 65, 194, 77, 250, 39 cleavage blastocyst rates establish dose. Results statistically analysed using Fisher’s exact test GraphPad Prism software (GraphPad Software Inc., La Jolla, CA, USA).
