ABSTRACT
The aim of this study was to evaluate the effect antioxidant dimethylthiourea (DMTU) as a supplement during collection and washing cumulus–oocyte complexes (COC) on apoptosis in cumulus cells (CC) competition for oocyte development vitro. COC were obtained by follicular aspiration. aspiration medium TCM-199 with 10% porcine fluid, 0.3 mM sodium pyruvate, penicillin/streptomycin. Treated groups supplemented 2 or 20 DMTU. After 44 h vitro maturation (IVM), percentage nuclear determined staining denuded oocytes Hoechst 33442 (n = 357). 18 fertilization (IVF) 15°C refrigerated spermatozoa from boars proven fertility, suspected zygotes stained 33342 percentages sperm penetration (SP), monospermic (MP), male pronucleus formation (MPN), IVF efficiency (monospermic oocytes/total oocytes) 260). In post-IVM CC, flow cytometry performed after annexin V-propidium iodide viability 90,000). Data analysed Flowing V 2.5.1 (http://flowingsoftware.btk.fi/). A comparison proportion among categories used (Fisher’s exact test), significant differences at P < 0.05. No effects observed maturation. Treatments DMTU significantly increased SP (Control: 35%; mM: 72.9%; 67.8%) without modifying MP. Although both treatments, amount MPN always greater than control, did not show differences. treatment (control: 22.2%; 31.9%; 45.7%). CC treated groups, increase 7.4%; 12.2%; 14.7%) decrease early 60.4%; 55.5%; 57.1%) observed. Late decreased (27.3%) compared other experimental 31.3%; 31.2%). addition reduce ROS-mediated optimize IVF. Besides, quality reflects oocytes, so analysis constitutes an interesting noninvasive method assess competence.
