ABSTRACT
The developing science called structural genomics has focused to date mainly on high-throughput expression of individual proteins, followed by their purification and structure determination. In contrast, the term biology is used denote determination structures, often complexes several macromolecules, that illuminate aspects biological function. Here we bridge with a procedure for determining protein previously unknown function from any organism sequenced genome. From computational genomic analysis, identify functionally linked proteins verify interaction in vitro coexpression/copurification. We illustrate this complex between PE PPE Mycobacterium tuberculosis genome, members families constitute ≈10% coding capacity predicted was readily expressed, purified, crystallized, although had failed expressing own. reason failure clear structure, which shows mate along an extended apolar interface form four-α-helical bundle, where two α-helices are contributed protein. Our entire identification, characterization, can be scaled genome-wide level.
