ABSTRACT
The release of mitochondrial intermembrane space proteins to the cytosol is a key event during apoptosis. We used in situ fluorescent labeling tagged with short tetracysteine-containing sequence follow Smac, Omi, adenylate kinase-2, cytochrome c, and apoptosis-inducing factor (AIF) apoptosis compared that c GFP individual cells observed over time. caspase-independent, simultaneous kinase-2. Although AIF also was caspase-independent commenced other proteins, it proceeded much more slowly incompletely from mitochondria, perhaps because requirement for secondary event. These results suggest these are released through same pore may not be regulated selective proteins. timing extent makes unlikely involved induction apoptosis, either upstream or downstream outer membrane permeabilization.
