Modulation of human natural killer T cell ligands on TLR-mediated antigen-presenting cell activation

ABSTRACT
Invariant natural killer T (iNKT) cells are a subset of nonconventional recognizing endogenous and/or exogenous glycolipid antigens in the context CD1d molecules. It remains unclear whether innate stimuli can modify profile lipids recognized by iNKT on surface antigen-presenting (APCs). We report that activation human APCs Toll-like receptor ligands (TLR-L) modulates lipid biosynthetic pathway, resulting enhanced recognition CD1d-associated cells, as defined IFN-gamma secretion. APC-derived soluble factors further increase CD1d-restricted cell activation. Finally, using tetrameric receptors (TCR) staining reagent, we demonstrate specific up-regulation CD1d-bound ligand(s) TLR-mediated APC maturation. The ability to modulate and maturation underscores role assisting priming antigen-specific immune responses.
