ABSTRACT
Significance Extensive studies have detailed the molecular regulation of individual components hemostatic system. Important questions remain, however, regarding how elements coagulation cascade, platelets, and vessel wall are integrated at systems level to achieve hemostasis in vivo. Here, a series high-resolution imaging studies, we explore detail plug composed platelets fibrin forms setting penetrating vascular injuries. The results reveal surprisingly heterogeneous architecture, including gradients platelet activation spatial sequestration thrombin activity outside lumen. They also show widely prescribed class antiplatelet agents alters resulting adverse bleeding certain physiologic contexts.
