ABSTRACT
CCAAT/enhancer binding protein (C/EBP) ɛ is a recently cloned member of the C/EBP family transcription factors and expressed exclusively in cells hematopoietic origin. The human C/EBPɛ gene transcribed by two alternative promoters, Pα Pβ. A combination differential splicing use promoters generates four mRNA isoforms, 2.6 kb 1.3–1.5 size. These transcripts can encode three proteins calculated molecular mass 32.2 kDa, 27.8 14.3 kDa. Accordingly, Western blots with antibodies specific for DNA-binding domain, that common to all forms, identify multiple proteins. was greatly induced during vitro granulocytic differentiation primary CD34 + cells. Retinoic acid treatment HL60 promyelocytic leukemia 24 hr levels 4-fold, while prolonged gradually reduced expression pretreatment levels. Transient transfection experiments vectors isoforms demonstrated 32.2-kDa an activator granulocyte colony-stimulating factor receptor promoter, 14.3-kDa not. Thus, regulated complex fashion may play role regulation genes involved myeloid differentiation.
