ABSTRACT
We have examined the role played by protein kinase A (PKA) in vesicle-mediated transport from trans-Golgi network (TGN) to cell surface. In vivo this step was inhibited inhibitors of PKA catalytic subunits (C-PKA) such as compound known H89 and a myristoylated form inhibitory peptide sequence contained thermostable inhibitor. Inhibition occurred at an early stage during transfer vesicular stomatitis virus G glycoprotein TGN Reversal inhibition correlated with transient increase number free coated vesicles Golgi area. Vesicle budding studied vitro using virus-infected, permeabilized cells. Addition assay C-PKA stimulated vesicle release while it suppressed peptide, H89, antibody against C-PKA.
