ABSTRACT
G 1 /S and 2 /M cell cycle checkpoints maintain genomic stability in eukaryotes response to genotoxic stress. We report here both genetic functional evidence of a Gadd45-mediated checkpoint human murine cells. Increased expression Gadd45 via microinjection an vector into primary fibroblasts arrests the cells at boundary with phenotype MPM2 immunopositivity, 4 n DNA content and, 15% cells, centrosome separation. The arrest depends on wild-type p53, because no was observed either p53-null Li–Fraumeni or normal coexpressed p53 mutants. cyclin B1 Cdc25C inhibited fibroblasts, indicating that mechanism is least part through modulation activity -specific kinase, B1/p34 cdc2 . Genetic physiological obtained by using GADD45 -deficient Human endogenous reduced antisense have impaired after exposure ultraviolet radiation methyl methanesulfonate but are still able undergo ionizing radiation. Lymphocytes from gadd45 -knockout mice ( −/−) also retained initiated failed Therefore, mammalian genome protected multiplicity specific types damage.
