ABSTRACT
The galectins are a family of β-galactoside-binding proteins implicated in modulating cell-cell and cell-matrix interactions. Here we report the cloning expression novel member this (galectin-7) that correspond to IEF (isoelectric focusing) 17 (12,700 Da; pI, 7.6) human keratinocyte protein data base, is strikingly down-regulated SV40 transformed keratinocytes (K14). cDNA was cloned from λgt11 library using degenerated oligodeoxyribonucleotides back-translated an peptide sequence. encoded by galectin-7 clone comigrated with as determined two-dimensional (two-dimensional gel electrophoresis) analysis expressed transiently transfected COS-1 cells, bound lactose. Alignment amino acid sequences other members showed acids central β-galactoside interaction conserved. Galectin-7 partially externalized medium although it has no typical secretion signal peptide. Immunoblotting well immunofluorescence tissues specific antibody revealed narrow distribution which found mainly stratified squamous epithelium. antigen localized basal keratinocytes, also found, albeit at lower levels, suprabasal layers where concentrated areas cell contact. Both, its cellular localization striking down-regulation K14 imply role and/or interactions necessary for normal growth control. gene mapped chromosome 19.
