ABSTRACT
To study the organization of DNA replication in mammalian rRNA genes, sites initiation synthesis rat and human genes were mapped by two independent techniques. In cells growth nascent chains was blocked Trioxsalen cross-links introduced vivo. The fraction “restricted” labeled vivo isolated, abundance this cloned ribosomal sequences determined hybridization. experiments with cells, allowed to grow unrestricted for a certain period time movement forks along followed hybridization “unrestricted” fragments fractionated according size. results show that both there are well defined regions synthesis. first one is located upstream transcription units second at 3′-end coding repeats.
