ABSTRACT
The serum and glucocorticoid-inducible kinase SGK1 increases the activity of Orai1, pore forming unit store-operated Ca(2+) entry, thus influences Ca(2+)-dependent cellular functions such as migration. further regulates transcription factor nuclear κB (NF-κB). This study explored whether Orai1 and/or STIM1, Orai1-activating sensor. STIM1 transcript levels were decreased in mast cells from knock-out mice increased HEK293 transfected with active (S422D)SGK1 but not inactive (K127N)SGK1 or (S422D)SGK1-transfected treated NF-κB inhibitor Wogonin (100 μm). Treatment stem cell enhanced sgk1(+/+) sgk1(-/-) Wogonin. by transfection subunit p65 well subunits p65/p50 p65/p52. They silencing p65, p50, p52 Luciferase assay chromatin immunoprecipitation defined NF-κB-binding sites promoter regions accounting for sensitive genomic regulation Orai1. Store-operated entry was similarly overexpression p65/p52 treatment Transfection augmented present observations reveal powerful Orai1/STIM1 SGK1-dependent signaling.
