ABSTRACT
The present study was performed to identify cells responsible for the elimination of T reactive with minor lymphocyte-stimulating (Mls) antigens during cell development. Experiments were carried out in a fetal thymus organ culture (FTOC) system. To examine tolerance-inducing activity, various populations from adult CBA/J (Mls-1a) mice injected into deoxyguanosine (dGuo)-treated FTOC C3H/He (Mls-1b) microinjector, and 2 d later, lobes as precursors. After 14 cultivation, harvested assayed expression receptor V beta 6 element. absence or marked reduction expressing at high levels (V 6high) regarded indicating deletion Mls-1a-reactive cells. cell-depleted thymic well splenic able induce clonal deletion. Further characterization effector by fractionating spleen before injecting them dGuo-FTOC. None dish-adherent population, dish-nonadherent purified B alone deletion, whereas addition adherent restored tolerance inducibility. It further shown that combination dendritic effective eliminating Mls-reactive clones. These results indicate clones Mls development thymus, both DC are required.
