Subsets of Human Dendritic Cell Precursors Express Different Toll-like Receptors and Respond to Different Microbial Antigens

ABSTRACT
Toll-like receptors (TLRs) are ancient microbial pattern recognition highly conserved from Drosophila to humans. To investigate if subsets of human dendritic cell precursors (pre-DC), including monocytes (pre-DC1), plasmacytoid DC (pre-DC2), and CD11c+ immature DCs (imDCs) developed recognize different microbes or antigens, we studied their TLR expression responses antigens. We demonstrate that whereas preferentially express 1, 2, 4, 5, 8, pre-DC strongly 7 9. In accordance with these profiles, respond the known ligands for TLR2 (peptidoglycan [PGN], lipoteichoic acid) TLR4 (lipopolysaccharide), by producing tumor necrosis factor (TNF)-α interleukin (IL)-6. contrast, pre-DCs only TLR9-ligand, CpG-ODNs (oligodeoxynucleotides [ODNs] containing unmethylated CpG motifs), IFN-α. imDCs 3 2-ligand PGN large amounts TNF-α, viral double-stranded RNA-like molecule poly I:C, IFN-α IL-12.
