Identification of Splice Variants, Targeted MicroRNAs and Functional Single Nucleotide Polymorphisms of the BOLA-DQA2 Gene in Dairy Cattle

ABSTRACT
Major histocompatibility complex, class II, DQ alpha 2, also named BOLA-DQA2, belongs to the Bovine Leukocyte Antigen (BOLA) II genes which are involved in immune response. To explore variability of BOLA-DQA2 gene and resistance mastitis cows, splice variants (SV), targeted microRNAs (miRNAs), single nucleotide polymorphisms (SNPs) were identified this study. A new SV (BOLA-DQA2-SV1) lacking part exon 3 (195 bp) two 3′-untranslated regions (UTR) (52 bp+167 was found healthy mastitis-infected mammary gland tissues. Four 13 SNPs multiple resulted amino acid changes protein SNP (c. +1283 C>T) may affect binding seed sequence bta-miR-2318. Further, we detected relative expressions transcripts five candidated 3′-UTR tissues dairy cattle by using quantitative real-time polymerase chain reaction. The result showed that expression BOLA-DQA2-SV1 mRNA significantly upregulated 2.67-fold (p<0.05) (n=5) compared with (n=5). Except for bta-miR-1777a, miRNA (bta-miR-296, miR-2430, miR-671) 1.75 2.59-fold (p<0.05), whereas miR-2318 downregulated cows. Our findings reveal play an important role cattle. Whether structure or association is unknown warrants further investigation.
