ABSTRACT
Assembly of the nuclear pore, gateway to genome, from its component subunits is a complex process. In higher eukaryotes, pore assembly begins with binding ELYS/MEL-28 chromatin and recruitment large critical Nup107-160 subunit. The choreography steps that follow largely speculative. Here, we set out molecularly define early in assembly, beginning binding. Point mutation analysis indicates exquisitely sensitive change only two amino acids AT-hook motif ELYS. dependence on AT-rich for ELYS borne by use DNA-binding antibiotics. AT-binding Distamycin A blocks whereas GC-binding Chromomycin A(3) does not. Next, find vesicles containing key integral membrane proteins POM121 NDC1 forming nucleus dependent chromatin-bound ELYS/Nup107-160 complex, gp210 Indeed, reveal an interaction between cytoplasmic domain complex. Our data thus suggest order 1) sites, 2) ELYS, 3) 4) POM121- NDC1-containing and/or sheets, followed (5) bulk remaining soluble subunits.
