ABSTRACT
: Diffusion maps are a spectral method for non-linear dimension reduction and have recently been adapted the visualization of single-cell expression data. Here we present destiny, an efficient R implementation diffusion map algorithm. Our package includes specific noise model allowing missing censored values. In contrast to previous implementations, further nearest-neighbour approximation that allows processing hundreds thousands cells functionality projecting new data on existing maps. We exemplarily apply destiny recent time-resolved mass cytometry dataset cellular reprogramming.destiny is open-source R/Bioconductor "bioconductor.org/packages/destiny" also available at www.helmholtz-muenchen.de/icb/destiny A detailed vignette describing functions workflows provided with package.carsten.marr@helmholtz-muenchen.de or f.buettner@helmholtz-muenchen.deSupplementary Bioinformatics online.
