ABSTRACT
Determining the methylation state of regions with high copy numbers is challenging for second-generation sequencing, because read length insufficient to map reads uniquely, especially when repetitive are long and nearly identical each other. Single-molecule real-time (SMRT) sequencing a promising method observing such regions, it not vulnerable GC bias, produces lengths, its kinetic information sensitive DNA modifications.
