Simultaneous Determination of Human Plasma Levels of Citalopram, Paroxetine, Sertraline, and Their Metabolites by Gas Chromatography--Mass Spectrometry

ABSTRACT
A gas chromatography-mass spectrometry method is presented which allows the simultaneous determination of plasma concentrations selective serotonin reuptake inhibitors citalopram, paroxetine, sertraline, and their pharmacologically active N-demethylated metabolites (desmethylcitalopram, didesmethylcitalopram, desmethylsertraline) after derivatization with reagent N-methyl-bis(trifluoroacetamide). No interferences from endogenous compounds are observed following extraction samples six different human subjects. The standard curves linear over a working range 10-500 ng/mL for 10-300 desmethylcitalopram, 5-60 20-400 sertraline desmethylsertraline, 10-200 paroxetine. Recoveries measured at three 81 to 118% tertiary amines (citalopram internal methylmaprotiline), 73 95% secondary paroxetine sertraline), 39 66% primary (didesmethylcitalopram desmethylsertraline). Intra- interday coefficients variation determined 3 11% citalopram its metabolites, 4 15% 5 13% desmethylsertraline. limits quantitation 2 1 0.5 noted 20 other psychotropic drugs.
