ABSTRACT
The ability of the Pseudomonas syringae pv. phaseolicola harpin (HrpZPsph) to elicit hypersensitive response was investigated in three Nicotiana genotypes. hrpZPsph gene placed under chemical regulation (tetracycline induction) TetR+ tabacum cv. Wisconsin 38 (W38) or transiently expressed N. benthamiana following infection with a PVX-derived vector and genotypes by agroinfiltration. constructs were designed express either canonical form an N-terminally extended version protein carrying signal peptide portion tobacco pathogenesis-related PR1a (SP-HrpZPsph). Stable transformants W38 did not develop necrosis upon induction tetracycline, probably as result insufficient accumulation. In contrast, plants infected PVX produced high concentrations biologically active form, but only those expressing secretable developed necrotic symptoms. These symptoms less severe than caused PVX::avrPto; however, they accompanied hsr203J, response-specific transcript. results suggest that plant cellular receptor(s) for is extracellular.
