ABSTRACT
A sensitive assay based on the polymerase chain reaction (PCR) has been developed to detect sugarcane bacilliform badnavirus (SCBV) in diseased plants. Two sets of primer pairs, which prime synthesis DNA from putative aspartic protease and reverse transcriptase coding regions, were evaluated. Best results obtained with pair SCBVF5 SCBVR5, primes a 221-bp virus-specific fragment total nucleic acid extracts The viral origin PCR products was confirmed by Southern hybridization 1,602-bp cloned SCBV genome. This diagnostic test used infection noble canes (Saccharum officinarum), commercial hybrids, clones within Saccharum complex including S. robusturn, spontaneum, barberi, Erianthus arundinaceus, E. ravennae, as well banana infected streak virus. screening show that is widely distributed throughout related germ plasm.
