ABSTRACT
In eastern Canada, soybean, Glycine max (L.) Merr., is the most important legume, and its cultivation expanding to new regions as cultivars for short growing season are developed. The soybean cyst nematode (SCN), Heterodera glycines Ichinohe, among destructive pests of in world. This also under quarantine regulations many countries, including Canada. Until now, SCN was only reported province Ontario. Since first detection 1988 southwestern part (1), has been found 12 other counties. It appears that spreading a north northeast direction along St. Lawrence River. We report here Quebec. Second stage juveniles (J2) cysts were Anicet, Quebec, 10-ha field. Light textured soil characteristic field, same site where Pratylenchus alleni recently discovered (2) irregular patches stunted plants observed. Morphological molecular studies J2 confirmed identification this population SCN. typical with body length 393 428 μm, lateral fields harboring four straight lines, well-developed stylet 23 25 μm long, sub-ventral base knobs posterior slops, tail 43 50 hyaline 29 μm. Cysts brown lemon-shaped protuberance, ambifenestrated, underbridged, had strongly developed bullae. Key morphometrics were: fenestra 40 57 long 28 44 wide, vulval slit 39 53 long. All these coincident those (3). Ribosomal DNA ITS, 18S, D2/D3 regions, mitochondrial COX1 gene PCR amplified from J2s gDNA using primers ITS-F (5'-TTGATTACGTCCCTGCCCTTT-3') ITS-R (5'-ACGAGCCGAGTGATCCACCG-3'); 18S-F (5'-TTGGATAACTGTGGTTTAACTAG-3') 18S-R (5'-ATTTCACCTCTCACGCAACA-3'); D2A (5'-ACAAGTACCGTGAGGGAAAGT-3') D3B (5'-GACCCGTCTTGAAACACGGA-3'); COXI-F (5'-CCTACTATGATTGGTGGTTTTGGTAATTG-3') COX1-R (5'-GTAGCAGCAGTAAAATAAGCACG-3'), respectively, sequenced. nucleotide sequences 98 100% similar NCBI nr database (July 2013). have submitted GenBank following accession numbers: ITS (KF453621); 18S (KF453622); (KF453623); (KF453624). Using species specific sequence characterized region (SCAR) (4) H. glycines. case proximity Anicet Ontario border accordance North/Northeastern dispersal hypothesis.
