ABSTRACT
Cardiovascular diseases are a leading cause of death and disability in diabetic patients. Recent studies suggest that switching gears towards survival longevity factors such as sirtuins induce protection against cardiovascular complications The present study investigated the effect SIRT1 activator II on myocardial injury high fat diet‐induced type 2 mice. Adult C57BL/6J mice (n=16/group) were fed normal chow diet (ND‐10 Kcal % fat) or high‐fat (HFD‐60 for 6 months. Body weights blood glucose levels monitored. Glucose tolerance tests performed after dietary intervention to confirm development diabetes. A subset HFD treated with Activator 4 weeks. Plasma was collected from measure plasma glucose, insulin lipid parameters total cholesterol, HDL LDL/vLDL. Subsequently hearts isolated subjected 20 min ischemia 40 reperfusion (I/R). treatment significantly reduced body weight (42.5±1.1 g vs 50.9±1.5 g; p<0.05), fasting (210.1±24.6 mg/dl 354.7±19.7 mg/dl; p<0.05) (3.97±1.45 ng/ml 12.11±2.01 ng/ml; p<0.02) compared untreated group. improved profile Functional recovery I/R ex vivo Langendorff perfusion model lower than ND mouse demonstrated better cardiac function had significant reduction necrotic markers creatine kinase lactate dehydrogenase. infarct size (22.08±2.2% 35.418±3.79% p<0.01) following injury. Additionally, suppressed expression proinflammatory cytokines In conclusion, our findings prevents detrimental effects diabetes could be safe therapeutic ischemic heart Support Funding Information DK104031
