ABSTRACT
Colorectal cancer (CRC) is a heterogeneous disease. Subtyping in gene expression recognized as relevant criterion for classification of the disease, but its clinical usefulness hampered by lack standard and methodology to be used between studies analyze responsiveness each patient chemotherapeutic treatment. CXCR4 (CD184) when highly expressed generates proliferation cell survival, angiogenesis metastatic tumor migration. When CD26 less expressed, there decrease local chemokine levels, thereby controlling situ. Patients responsive 5‐FU, FOLFOX FOLFIRI present significant reduction progressive increase CD26, where inverse ratio seen nonresponsive patients. Thus, order standardize high sensitivity diagnosis prognosis, we developed use technology detection circulating antigens through nanotechnology associated with antibodies. following vitro methodology, anti‐CXCR4 anti‐CD26 pAbs were due producing clones IgM or IgG subtypes marker, since difference expressions such genes suggests an interesting imbalance, generating possible response. After standardization induction nude mice, HT‐29 cells (mutation at codon 273 p53 gene) performed, four inoculations these proteins carried out throughout production process, immunoenzymatic assays performed identification selection Positive. The purified antibodies tested SDS‐PAGE Western Blotting confirmation specificity. Thereafter, removed from mice will labeled verify on their surfaces inoculated pAb thereafter, tissues patients different stages disease also labeled. We thus sought, cases, assist medical team decision treatment leading surgical intervention and/or conventional chemotherapies.
