The antitumoral effect of endostatin and angiostatin is associated with a down‐regulation of vascular endothelial growth factor expression in tumor cells

ABSTRACT
Endostatin and angiostatin are known as tumor‐derived angiogenesis inhibitors, but their mechanisms of action not yet completely defined. We report here that endostatin angiostatin, delivered by adenoviral vectors, reduced in vitro the neovessel formation mouse aortic ring assay 85 40%, respectively. also demonstrated vivo both inhibited local invasion tumor vascularization transplanted murine malignant keratinocytes, 50 90% development highly vascularized mammary tumors. This inhibition growth was associated with a reduction vascularization. Expression analysis vascular endothelial factor (VEGF) carried out model revealed 3‐ to 10‐fold down‐regulation VEGF mRNA expression endostatin‐treated rings. A similar at protein levels observed two cancer models after treatment each inhibitor. suggests effects may be mediated, least part, ability down‐regulate within tumor. work provides evidence act on cells themselves.
