ABSTRACT
Alterations in mitochondrial dynamics, including their intracellular trafficking, are common early manifestations of neuronal degeneration. However, current methodologies used to study trafficking events rely on parameters that primarily altered later stages neurodegeneration. Our objective was establish a reliable applied statistical analysis detect alterations trafficking. We propose novel quantitative mitochondria trajectories based innovative movement descriptors, straightness, efficiency, anisotropy, and kurtosis. evaluated time- dose-dependent trajectory descriptors using biological data from differentiated SH-SY5Y cells treated with the toxicants 6-hydroxydopamine rotenone. MitoTracker Red CMXRos-labelled analyzed by total internal reflection fluorescence microscopy followed computational modelling describe process.
