Choline or CDP-choline attenuates coagulation abnormalities and prevents the development of acute disseminated intravascular coagulation in dogs during endotoxemia

ABSTRACT
Sepsis/endotoxemia causes platelet dysfunctions, abnormalities in coagulation and hemostatic mechanisms leading to organ dysfunctions mortality. Choline prevents injury improves survival during endotoxemia. The main objective of the present study was determine effects choline or cytidine-5'-diphosphocholine (CDP-choline) on endotoxin-induced activation development disseminated intravascular (DIC). Dogs were treated intravenously (i.v.) with saline, (20 mg/kg), CDP-choline (70 mg/kg) three times 4-h intervals starting 5 min before i.v. injection endotoxin (1 mg/kg). Platelet counts functions, prothrombin time (PT), activated partial thromboplastin (aPTT), fibrinogen, factors, D-dimer antithrombin (AT) measured at 0.5-96 h after endotoxin. Circulating platelet, factors AT decreased, whereas PT aPTT prolonged serum levels elevated Endotoxin-induced reductions attenuated blocked by CDP-choline. prolongation enhancement D-dimer. Elevated DIC scores administration increased concentrations caused bradycardia. also acetylcholine contents circulating mononuclear cells inhibited radioligand binding their cholinergic receptors. These data show that administration, as chloride CDP-choline, restores primary, secondary, tertiary hemostasis experimental endotoxemia dogs probably increasing both neuronal non-neuronal activity.
