ABSTRACT
To investigate the role of sphingosine kinase 1 (SphK1)/sphingosine 1-phosphate (S1P) signaling in inflammatory response severe acute pancreatitis (SAP).SAP is an process pancreas, which may lead to systemic syndrome and multiorgan dysfunction syndrome. SphK1 its product S1P have been implicated various immune cell functions. However, potential for SphK1/S1P SAP still unclear.Twenty-two patients with were enrolled this study. expression on peripheral neutrophils, monocytes, lymphocytes was evaluated by flow cytometry. SphK enzymatic activity neutrophils measured using a radiometric assay. The S1P1 S1P3 mRNA determined reverse transcriptase-polymerase chain reaction (RT-PCR). serum levels tumor necrosis factor-α (TNF-α), interleukin-1 (IL-1β), IL-6 ELISA.The markedly increased cells early stage then reduced restoration patients. Moreover, we found that level significantly elevated as compared healthy volunteers, it period. human CD4(+) T positively correlated APACHE (Acute Physiological Chronic Health Evaluation) II scores SAP. proinflammatory cytokines including TNF-α, IL-1β, showed similar shifts intracellular patients. The authors identified link between severity Observations possible immunomodulating SAP, suggesting regulation pathway represent novel targets treatment
