Detection and antibiotic resistance of Salmonella isolates from selected poultry farms in Dar es Salaam, Tanzania

ABSTRACT
Introduction. Salmonella is one of the most prevalent foodborne bacteria, posing a significant global health concern and responsible for ~155,000 deaths 93.8 million human illnesses annually. The rampant use antibiotic agents to combat salmonellosis in poultry has contributed emergence resistance against commonly used antibiotics. Methodology. cross-sectional study was conducted between January June 2023 across three districts Dar es Salaam region, Tanzania. isolates were detected confirmed by using standard microbiological conventional methods molecular such as PCR genomic sequencing. detecting presence invA gene, partial DNA sequencing performed identify species their close relatedness. A Kirby–Bauer disc diffusion method employed evaluate sensitivity seven different antibiotics, namely ampicillin, azithromycin, chloramphenicol, ciprofloxacin, gentamycin, trimethoprim/sulfamethoxazole tetracycline. strain with reference number ATCC 8739 control. Results. overall from faecal droppings 6.04% ( n =796). Of PCR-confirmed isolates, 64.3% =28) resistant more than two classes antibiotics hence considered multidrug resistant. highest observed ampicillin (92.9%, =26), followed tetracycline (71.43%, =20), ciprofloxacin (42.9%, =12), sulphonamide (42.85%, gentamicin (35.7%, =10) azithromycin (28.9%, =8). All susceptible chloramphenicol (100%, =28). Twenty-eight sent sequencing, out which 16 sequences (OR021717-OR021739) met criteria phylogenetic analysis. had per cent identity EU348369 strains Senftenberg isolated China, OL581594 Newport China EU348368 Pullorum China. Other diverged distantly; these are Abony accession CP007541 Kentucky OL581592. tree also included an outgroup species, bongori , downloaded GenBank numbers NC015761 NC021870 S. . Conclusion. high level found this could be due misuse management, and/or, probably, there circulating environment. To reverse trend observed, immediate interventions advocating prudent production systems strengthening extension services farmers farmer field school model improve management through improved farm biosecurity required.
