ABSTRACT
The aim of this study was to develop an immunogenic protective product against Shigella flexneri by employing a simple and safe heat treatment-based strategy. The physicochemical characteristics naturally produced (OMV) heat-induced (HT) outer-membrane vesicles from S. were examined, including comparison the protein content products. Toxicological biodistribution studies, preliminary experiment examine effectiveness HT in murine model infection, also included. This method simultaneously achieves complete bacterial inactivation production vaccine product, leading working process. obtained complex presented similar morphology (electron microscopy) chemical composition classical OMV, although it enriched some immunogens, such as lipoproteins, OmpA or OmpC, among others. formulation not toxic studies performed mice demonstrated that remained small intestine after nasal administration. Finally, single dose administered nasally able protect 2a.The convenient manufacturing process, biological evaluation, support use self-adjuvanted new candidate face shigellosis. Further development is required, additional immune analyses, evaluate whether subunit can be useful achieving full protection Shigella.
