ABSTRACT
Background IgE antibodies specific for the major birch pollen allergen frequently cross-react with Bet v 1 homologous food proteins, example Cor a in hazelnut and Mal d apple. Specific immunotherapy (BP-SIT) induces IgG4 that inhibit binding to 1. However, information on cross-reactivity of BP-SIT-induced 1-specific allergens is limited. In this study, we investigated kinetics production, cross-reactivity, IgE-blocking activity emerging during conventional BP-SIT whether IgG4-epitopes overlapped epitopes. Methods levels 1, were determined 42 pollen–allergic patients before BP-SIT. Inhibition was studied by IgE-facilitated antigen-binding assays basophil activation tests. Furthermore, inhibition IgE-mediated allergen-reactive T-cell lines assessed. Competitive immunoscreening phage-displayed peptides applied select mimotopes recognized antibodies, respectively. The resulting mapped surface 3D structure using computer-based algorithm. Results significantly increased 1- antibodies. parallel, allergen-specific decreased significantly. Sera containing inhibited binding, activation, allergen-induced proliferation. Predicted epitopes all showed high overlap. Conclusion Our results indicate may induce related epitope competition.
