ABSTRACT
miR-10a-5p has been shown to regulate cancer cell proliferation and invasiveness endothelial inflammatory responses. The function of in the skin not previously studied. aim current study was examine expression, regulation, keratinocytes (KCs) association with atopic dermatitis (AD).The expression its target genes analyzed using RT-qPCR, mRNA array analysis, situ hybridization, immunofluorescence. transfection miRNA mimics, cycle distribution luciferase assays used functions human primary KCs.miR-10a-5p found be upregulated lesional from patients AD proliferating KCs. Array pathway analysis IL-1β-stimulated KCs revealed that inhibited many affect progression only a few inflammation-related genes. Accordingly, fewer cells S-phase reduced were detected as characteristics miR-10a-5p-transfected influence on also evident induced by AD-related cytokines, including IL-4, IL-17, IL-1β, measured capacity strongly suppress marker Ki-67. Among putative direct genes, we verified hyaluronan synthase 3, damage-associated positive regulator KC migration proliferation, miR-10a-5p.miR-10a-5p inhibits directly targets 3 thereby may modulate AD-associated processes skin.
