Abstract Kinesin family member 20B ( KIF 20B, also known as MPHOSPH 1) is a kinesin protein that plays critical role in cytokinesis. Previously, we and others have demonstrated the oncogenic of several cancers; however, exact mechanisms underlying its tumorigenic effects remain unclear. Herein, showed overexpression human hepatocellular carcinoma HCC ) reported negative correlation between level prognosis patients. Mechanistically, reducing blockades mitotic exit cells at telophase spindle assembly checkpoint independent way. Importantly, acts synergistically with three microtubule‐associated agents MTA to p53‐ or p14 ARF ‐dependently suppress p53‐wt p53‐null cells. In addition taxol, enhanced toxicity two chemotherapeutic drugs, hydroxycamptothecin mitomycin C. conclusion, found novel mechanism blocking cytokinesis by inhibition increases efficacy ; our results thus suggested dual‐mitotic suppression approach against combining inhibition, which simultaneously blocks mitosis both metaphase telophase.
