Pyroptotic macrophages promote proliferation and chemotherapy resistance of peripheral T‐cell lymphoma via TLR4 signaling pathway

ABSTRACT
Peripheral T-cell lymphoma (PTCL) is a highly aggressive type of non-Hodgkin's with poor prognosis. Pyroptosis newly discovered procedural cell death mode, which has been implicated to occur in both tumor cells and immune cells. However, the occurrence effect pyroptosis on PTCL remain unclear. Here, we found that occurred interstitial macrophages rather than In clinical specimens, macrophage was associated prognosis PTCL. vitro experiments gene sequencing results showed pyroptotic could upregulate expression TLR4 through secreting inflammatory cytokines IL-18. Upregulated activated its downstream NF-κB anti-apoptotic signaling pathway, thus leading malignant proliferation chemotherapy resistance Moreover, factors such as XIAP pathway downregulated after knockdown TLR4, promotion also reversed. Our findings revealed mechanism promoting biological behavior elucidated key role this process. In-depth analysis will contribute understanding regulatory by microenvironment providing new ideas for treatment
