ABSTRACT
Human CD46 is a receptor for the M protein of group A streptococcus (GAS). The emm1 GAS strain GAS472 was isolated from patient suffering streptococcal toxic shock-like syndrome. CD46-expressing transgenic (Tg) mice developed necrotizing fasciitis associated with osteoclast-mediated progressive and severe bone destruction in hind paws 3 days after subcutaneous infection 5 × 10(5) colony-forming units GAS472. induced expression activator nuclear factor-κB ligand (RANKL) while concomitantly reducing osteoprotegerin limb bones Tg mice. Micro-computed tomography analysis suggested that local erosion systemic loss Because treatment monoclonal antibodies (mAbs) against mouse CD4(+) CD8(+) T lymphocytes did not inhibit osteoclastogenesis, lymphocyte-derived RANKL considered major contributor to massive during infection. However, immunohistochemical showed stimulated production various marrow cells, including fibroblast-like cells. Treatment mAb significantly inhibited osteoclast formation resorption. These data suggest increased heterogeneous cells provoked
