ABSTRACT
Abstract Aims This study was designed to investigate the role of NR2B and contribution DNA methylation expression in pathogenesis PND. Methods Eighteen‐month‐old C57BL/6J mice were subjected experimental laparotomy under 1.4% isoflurane anesthesia. Hippocampus‐dependent learning memory evaluated by using Barnes maze contextual fear conditioning tests. The protein mRNA levels western blotting qRT–PCR respectively, gene examined targeted bisulfite sequencing. Long‐term synaptic plasticity (LTP) measured electrophysiology. Results Mice that underwent exhibited hippocampus‐dependent cognitive deficits accompanied decreased expressions LTP deficiency. overexpression dorsal hippocampus could improve laparotomy. In particular, induced attributed hypermethylation. Preoperative administration S‐adenosylmethionine (SAM) hypomethylate gene, upregulate LTP, exerting a dose‐dependent therapeutic effect against Moreover, inhibiting abrogated benefits SAM pretreatment. Conclusions Laparotomy cause decline hypermethylating allowing us understand PND an epigenetic landscape.
