ABSTRACT
Abstract Background Pharmacokinetic (PK) studies of low‐dose prophylaxis (LDP) coagulation factor VIII (FVIII) in children with severe haemophilia A (SHA) are scarce. Objective This study aims to investigate the PK profile SHA receiving LDP FVIII. Methods Paediatric patients FVIII infusions (10 IU/kg twice weekly) were included. profiles estimated using Web Accessible Population Service for Haemophilia (WAPPS‐Haemo). The primary outcomes terminal half‐life (t 1/2 ), concentration–time profile, and time reach an level < 1%. secondary outcome was suggested dosing interval based on individual profile. Results Twenty‐five recruited; their mean age 12.3 ± 3.0 years. t differed among weekly, a median 14.8 h (IQR 12.6–16). 1% 73.8 58.8–80.3). Most could maintain trough > longer than 48 h. At 72–96 h, needed second dose infusion because ranged from daily every 96 depending Conclusion Our identified inter‐individual differences parameters weekly. results different intervals advise timing LDP. Estimating enables identification optimal frequency prevent bleedings.
